Introduction
GalNAc nucleic acid conjugates are divided into three parts: a three contact GalNAc target, a connecting arm, and a small nucleic acid molecule. GalNAc is covalently conjugated to the 3 'end of the sense chain of siRNA of different sequences in a trivalent state, forming polysaccharide siRNA single conjugates for direct and targeted delivery.
Delivery principle
GalNAc is a ligand for the asialoglycoprotein receptor (ASGPR), which can specifically bind to it. ASGPR and clathrin mediated endocytosis can effectively transport it to the cytoplasm, achieving rapid cellular endocytosis.
